| dc.description | Immature embryo culture for rapid fixation of inbred lines is of particular interest if selection is possible at the same time. A methodology is presented for determining downy mildew resistance in plants from immature embryos. All concentrations between 5000 and 500 000 zoosporangia/ml gave infection levels close to 100%. Seedling age was important. At the cotyledon stage 3% of plants escaped infection, while with 1-2 pairs of true leaves 99.5% of plants were infected. Passage in a humid chamber for 48 h promoted infection. The best method was to spray plants having one pair of true leaves with a suspension of 500 000 zoosporangia/ml and keep them for 48 h in a saturated atmosphere. A breeding programme is proposed to introduce downy mildew resistance and cytoplasmic male sterility using in vitro culture. | |
